      BackgroundThe translocase of outer mitochondrial membrane (TOM) complex is a multisubunit complex involved in the recognition, unfolding, and translocation of preproteins into the mitochondria. TOM20 and TOM70 are the two major receptor subunits of the TOM complex and constitute the binding site for mitochondrial proteins. TOM70 is anchored in the outer membrane by a hydrophobic segment near the N-terminus with the 65 kD C-terminus facing the cytosol. TOM70 forms a dimer and is associated with the TOM complex via interactions with TOM20. TOM70 binds various precursor proteins, including cytochrome c1 and aldehyde dehydrogenase III. Substrates for TOM70 are highly hydrophobic proteins and require molecular chaperones in the cytosol, such as Hsp70 and mitochondrial import stimulating factor (MSF), to maintain themselves in an import-competent state. Phosphorylation of TOM70 has been detected in screens of the mitocohondrial proteome and TOM70 had been shown to be required for proper targeting of the apopoptsis regulator Mcl-1.
