      Background InformationRAD50 (previously known as DNA repair protein RAD50) is homologous to Saccharomyces cerevisiae Rad50 and is a member of the structural maintenance of chromosome (SMC) family and is involved in cell growth and viability. RAD50 is known to encode 2 distinct proteins. The RAD50, XRS2/NBS1 complex is required for activation of ATM which will phosphorylate TERF1, thereby promoting telomerase. Along with MRE11 and NBN, RAD50 also forms the MRE11–RAD50–NBS1 (MRN) complex, which is one of the first factors to be localized to the DNA lesion, where it might initially have a structural role by tethering together, and therefore stabilizing, broken chromosomes. MRN also controls ATR, which is required for SMC1A phosphorylation, resulting in chromatid adhesion and DNA damage response. RAD50 is expressed in all tissue but predominantly in the testis and fibroblasts.

      UniProt SummaryFUNCTION:Component of the MRN complex, which plays a central role in double-strand break (DSB) repair, DNA recombination, maintenance of telomere integrity and meiosis. The complex possesses single-strand endonuclease activity and double-strand-specific 3'-5' exonuclease activity, which are provided by MRE11A. RAD50 may be required to bind DNA ends and hold them in close proximity. This could facilitate searches for short or long regions of sequence homology in the recombining DNA templates, and may also stimulate the activity of DNA ligases and/or restrict the nuclease activity of MRE11A to prevent nucleolytic degradation past a given point.       Storage and Shipping Information
      Storage ConditionsStable for 1 year at 2-8ºC from date of receipt.
      To reconstitute the antibody, centrifuge the antibody at a moderate speed (5,000 rpm) for 5 minutes. Carefully remove the ammonium sulfate/PBS buffer solution and discard; 10μL of residual ammonium sulfate solution will not affect the re-suspension of the antibody. Do not let the protein pellet dry, as severe loss of antibody reactivity can occur. Re-suspend the antibody pellet in 100L either standard PBS or TBS (pH 7.3-7.5). DO NOT VORTEX. Mix by gentle stirring with a wide pipet tip or gentle finger-tapping. Let the precipitated antibody rehydrate for 1 hour at 4-25°C prior to use. Small particles of precipitated antibody that fail to re-suspend are normal. Vials are overfilled to compensate for any losses.
