Application NotesParaffin Sections (1-2 µg/ml, trypsin and heat pre-treatment required)
Application CommentsStaining of formalin-fixed tissues requires digesting sections with trypsin at 1 mg/ml PBS for 10 min at 37°C, followed by boiling tissue sections in 10 mM citrate buffer, pH 6.0, for 10-20 min followed by cooling at RT for 20 min. Variables associated with assay conditions will dictate the proper working dilution.

ApplicationParaffin Sections (1-2 µg/ml, trypsin and heat pre-treatment required)
DescriptionProtein G purified mouse monoclonal antibody. Recognizes the ~205 kDa myosin heavy chain-1 and the ~200 kDa myosin heavy chain-2.
BackgroundSmooth muscle is an important element of the vascular, digestive, urogenital and respiratory systems. Smooth muscle cells (SMCs) are spindle-shaped cells that contain a variety of obliquely arrayed actomyosin-rich contractile elements that are attached to the plasma membrane. Smooth muscle heavy chain (SM-MHC) is a structural protein that is found in the cytoplasm. SM-MHC is a major component of the contractile apparatus in smooth muscle cells. Alternative splicing of the primary transcript from a single gene generates four smooth muscle myosin heavy chain isoforms. These isoforms differ both at the carboxyl terminus (SM1 and SM2 isoforms) and at the amino terminus (SM-A and SM-B isoforms). The smooth muscle myosin heavy chain isoforms are differentially expressed during smooth muscle development and in different smooth muscle cell types.
