      DescriptionAnti-CD35 Antibody, clone E11
      Alternate Names
      • Complement receptor type 1
      • C3b/C4b receptor
Background InformationComplement receptor type 1 (UniProt: P17927; also known as C3b/C4b receptor, CD35) is encoded by the CR1 (also known as C3BR) gene (Gene ID: 1378) in human. CD35 is a single-pass type I membrane glycoprotein of the receptors of complement activation (RCA) family. It is synthesized with a signal peptide (aa 1-42), which is subsequently cleaved off to generate the mature form that contains a long extracellular domain (aa 42-1971) , a transmembrane domain (aa 1972-1996), and a cytoplasmic domain (aa 1997-2039). It contains 30 Sushi domains. Its expression has been reported on erythrocytes, a subset of T-cells, mature B cells, follicular dendritic cells, monocytes, and granulocytes. It serves as a membrane immune adherence receptor that plays a critical role in the capture and clearance of complement -opsonized pathogens by erythrocytes and monocytes/macrophages. Its ligands include C3b, iC3b, and C4b fragments of C3 and C4 proteins. CD35 also acts as an inhibitor of spontaneous complement activation by impairing the formation and function of the alternative and classical pathways C3/C5 convertases, and by serving as a cofactor for the cleavage of C3b to iC3b, C3c, and C3d, C3g, and C4b, C4c, and C4d by factor I. It also plays a role in immune regulation by contributing, upon ligand binding, to the generation of regulatory T cells from activated helper T cells and inn maintaining a balance between normal and pathological B-cell activation.
