      Background InformationAllergin-1 (UniProt: Q3TB92; also known as Allergy inhibitory receptor 1, Mast cell antigen 32, MCA-32, Mast cell Ag-32, Mast cell immunoglobulin-like receptor 1) is encoded by the Milr1 (also known as Gm885, Mca32) gene (Gene ID: 380732) in murine species. Allergin-1 is a single-pass type I membrane protein that is expressed in mast cells, dendritic cells, macrophages and neutrophils, but not in T cells, B cells, or natural killer cells. myeloid cells. It serves as an immunoglobulin-like receptor and plays an inhibitory role in degranulation of mast cells. Also, it negatively regulates IgE-mediated mast cell activation and suppresses the type I immediate hypersensitivity reaction. Allergin-1 is reported to recruit SHP-1 and SHP-2, which inhibits high-affinity receptors involved in IgE (Fc RI)-mediated signaling and subsequent mast cell activation. Allergin-1 is shown to suppress IgE-mediated, mast cell dependent anaphylaxis in mice and mice deficient in Allergin-1 develop enhanced passive systemic and cutaneous anaphylaxis. Allergin-1 is synthesized with a signal peptide (aa 1-33), which is subsequently cleaved off to produce the mature form that contains an extracellular domain (aa 34-150), a transmembrane domain (aa 151-171), and a cytoplasmic domain (aa 172-246). Mouse Allergin-1 contains two ITIM motifs in its cytoplasmic domain (aa 214-219 and 239-244). Two isoforms of murine Allergin-1 have been described that are produced by alternative splicing. Clone TX83 was generated by immunizing Allergin-1 deficient mice with BW5147 cells expressing FLAG-tagged Allergin-1. (Ref.: Tsurusaki, S., et al. (2016). Int.
