      Overview

      This product has been discontinued.

Has been validated for use as a positive control for protein tyrosine kinase (PTK) activity assays. There is sufficient PTK activity at a concentration of 10-20 ng/well using the K-LISA™ PTK Screening Kit (Cat. No. 539701).
      Catalogue Number325540
      Brand Family Calbiochem®
      Application Data
The rat spleen extract was reconstituted as indicated above, diluted in reaction buffer, and assayed using the K-LISA™ PTK Screening Kit according to the recommended protocol. Absorbance values shown represent the mean of triplicate samples that were corrected against background absorbance. Background absorbance values ranged from ~0.05-0.07 absorbance units.

DescriptionThe Protein Tyrosine Kinase Positive Control, Rat Spleen Extract, 7 mg has been validated for use as a positive control for protein tyrosine kinase (PTK) activity assays. Assay Characteristics and Examples

The assay example below represents typical data obtained using the rat spleen extract in a tyrosine kinase assay. Actual tyrosine kinase activity may vary from lot to lot.
Recommended reaction conditionsReagent Preparation Reconstitute the entire vial of rat spleen extract with 250 µl of ultra pure H2O. The resulting suspension has a concentration of 28 mg/ml total protein. Dispense into aliquots and store at -70°C. Use one aliquot as positive control for tyrosine kinase assays. There is sufficient PTK activity at 10-20 ng/well to serve as a strong positive control (see example below). Discard the unused portion; do not re-freeze.
