      Full-length, recombinant, human phospholipase C γ-1 fused at the N-terminus to His•Tag® and S•Tag™ sequences and expressed in S. frugiperda insect cells. This preparation is qualified for use as a substrate for protein tyrosine kinases in in vitro assays. Intrinsic enzyme activity has not been tested. PLC-γ-1 is a major substrate for heparin-binding growth factor 1-activated tyrosine kinase. Receptor-mediated activation of PLC-γ-1 and PLC-γ-2 involves their phosphorylation by tyrosine kinases in response to ligation of a variety of growth factor receptors and immune system receptors. The full-length protein contains the following domains: C2 domain (aa 1075-1177); EF-hand calcium-binding domain (aa 165-176); two PH domains (aa 27-142, aa 4889-523 and aa 895-931); PI-PLC X-box catalytic domain (aa 320-464); PI-PLC Y-box catalytic domain (aa 953-1070); two SH2 domains (aa 550-657, aa 668-756); and a SH3 domain (aa 791-851). Phosphorylation is known to occur at Tyr771 and Tyr1253 by EGFR, Tyr783 by Syk, Ser1248 by PKC-α, and Tyr432 and Tyr775 probably by FGFR.
