      Background InformationThe N2A Psen1/Psen2 cell line is a model for studies of presenilin function in the pathology of familial Alzheimer’s Disease. Early-onset Alzheimer’s disease (AD), also termed autosomal dominant AD with early onset (ADAD), is a devastating neurodegenerative disease that is strongly linked to familial inheritance. The main cause of early-onset/familial AD is autosomal dominant mutations in APP (amyloid precursor protein), PSEN1 (presenilin-1) or PSEN2 (presenilin-2).1 As catalytic subunits of γ-secretase complex, which cleaves APP to form amyloid beta peptides, Psen1 and Psen2 play significant roles in the formation of amyloid plaques. Variations in PSEN1/2 are frequently present in familial early-onset AD cases but their functional significance is unknown. Unraveling the functions of presenilin proteins is thus key to understanding the molecular pathology of AD.

      The N2A Psen1/2 cell line was derived via CRISPR/Cas9-editing of the parental N2A mouse neuroblastoma cell line.2 The N2A (Neuro2a) parental cell line was derived from neuroblastoma stem cells of a male strain A/J mouse.4
