      DescriptionPurified mouse monoclonal antibody generated by immunizing BALB/c mice with the specified immunogen and fusing splenocytes with P3X63Ag8.653 mouse myeloma cells. Recognizes the ~170 kDa topoisomerase II protein.
      BackgroundTopoisomerase II is a nuclear enzyme whose primary function is to induce temporary double-strand breaks in DNA and to facilitate the passage of the duplex DNA segments through these breaks. Phosphorylation of topoisomerase II in a cell cycle regulated manner appears to be important for progression through the cell cycle. The enzyme exists as two discrete isozymes, topoisomerase IIα (~170 kDa) and topoisomerase IIβ (~180 kDa) which are under independent transcriptional regulation. Both isozymes have been identified in human and mouse cells but only the topoisomerase IIα form appears in yeast. The gene encoding topoisomerase IIα has been mapped to chromosome 17 and is in close proximity to the gene for erbB-2. Altered expression of topoisomerase II has been documented in many cancers including AML, CLL, small cell lung cancer, breast and ovarian cancer, and colon cancer. Based on this observation, topoisomerase II has been identified as the target of many widely used antineoplastic drugs as well as the target of rational drug design.
