      Background InformationAspartate aminotransferase, cytoplasmic (EC 2.6.2.1.; EC 2.6.2.3; UniProt P17174; also known as Aspartate aminotransferase 1, Aspartate transaminase 1, cAspAT, cCAT, Cysteine aminotransferase cytoplasmic, Cysteine transaminase cytoplasmic, Glutamate oxaloacetate transaminase 1, Growth-inhibiting protein 18, Transaminase A) is encoded by the GOT1 (also known as GIG18, ASTQTL1) gene (Gene ID 2805) in human. Aspartate aminotransferase is expressed in many types of cells and tissues and is conserved among prokaryotic and eukaryotic organisms. There exist two highly similar AST forms in mammals, the cytosolic cAST (encoded by the GOT1 gene) and the mitochondrial mAST (encoded by the GOT2 gene). Serum aspartate aminotransferase activity is largely that of cAST/GOT1, and elevated serum AST activity is associated with liver, heart, and muscle injury. AST catalyzes the reversible conversion between aspartate/alpha-ketoglutarate and oxaloacetate/glutamate, increasing oxaloacetate concentration therefore shifts the equilibrium of AST-catalzed reaction toward aspartate/alpha-ketoglutarate and away from glutatmate. Lowering blood glutamate concentration via oxaloacetate i.v. injection prompts an efflux of excess extracellular brain glutamate into the blood, which is reported to allevate the infarct size and edema in a rat model of ischemia induced by transient occlusion of the middle cerebral artery occlusion (MCAO).
