      Background InformationFolate receptor alpha (UniProt: P15328; also known as FR-alpha, Adult folate-binding protein, FBP, Folate receptor 1, Folate receptor, adult, KB cells FBP, Ovarian tumor-associated antigen MOv18) is encoded by the FOLR1 (also known as FOLR) gene (Gene ID: 2348) in human. Folate receptors exist in three major forms, namely FR-alpha (FOLR1), FR-beta (FOLR2), and FR-gamma (FOLR3) and the alpha isoform is reported to be over-expressed in several tumors, including ovarian, endometrial, colorectal, breast cancers. FOLR1 is primarily expressed in tissues of epithelial origin. It binds to folate and reduced folic acid derivatives and mediate the delivery of 5-ethyltetrahydrofolate and folate analogs into the interior of cells. It displays high affinity for folate and folic acid analogs at neutral pH. FOLR1 is synthesized with a signal peptide (aa 1-24) and a propeptide (aa 235-257) that are cleaved off in the mature form (aa 25-234). A secreted form of FORL1 has also been described that is derived from the membrane-bound form either by cleavage of the GPI anchor or via proteolysis catalyzed by a metalloprotease. Mutations in FOLR1 gene are known to cause neurodegeneration due to folate transport deficiency that leads to severe developmental regression, movement disturbances, epilepsy, and leukodystrophy.
