      Background InformationProgrammed cell death 6-interacting protein (UniProt: Q9WU78; also known as ALG-2-interacting protein 1, ALG-2-interacting protein X, E2F1-inducible protein, Eig2) is encoded by the Pdcd6ip (also known as Aip1, Alix) gene (Gene ID: 18571) in murine species. AIP1/ALIX is a class E VPS cytoplasmic, ubiquitously expressed protein that is involved in concentration and sorting of cargo proteins of the multi-vesicular body (MVB) for incorporation into intra-lumenal vesicles that are generated by invagination and scission from the limiting membrane of the endosome. MVB-mediated sorting is driven by five distinct ESCRT complexes (ESCRT-0, -I, -II and -III, and the Vps4 complex) and associated proteins. AIP1/ALIX and charged MVB protein 4 (CHMP4) are binding partners involved in a variety of ESCRT-III-mediated membrane-remodeling in mammalian cells. In the absence of binding partners, an intramolecular interaction between the Patch 2 region in the BRO1 domain (aa 3-392) and the TSG101-docking site (aa 717-720) in the Pro-rich domain (aa 717-860) locks ALIX in a conformation inaccessible for interaction with CHMP4 and retroviral Gag proteins. AIP1/ALIX interacts with ALG-2 (apoptosis-linked gene-2) to regulate cell death. This interaction is strictly dependent on calcium. It is also known to regulate exosome biogenesis in concert with SDC1/4 and SDCBP.
