Merck’s assay kits and antibodies (including: CoA, Lysine, Cortactin, p53, STAT5B and Prolactin Receptor). measure both acetylation and deacetylation activity, helping to elucidate the relationship between chromatin modification pathways and gene regulation.
The reversible acetylation of lysine amino groups in nuclear histones is an important mechanism of regulating gene expression. Acetylated histone proteins confer accessibility of the DNA template to the transcriptional machinery for expression. Histone acetyltransferase enzymes (HATs) preferentially acetylate specific histone lysine substrates. Histone acetylation has been linked to gene-specific activation by transcription factors. It plays an important role in cell cycle control and has been linked to the uncontrolled cell proliferation. Histone hyperacetylation is well correlated with increased transcription, whereas histone deacetylation (by histone deacetylases, or HDACs) correlates with transcriptional repression.

Anti-acetyl BMAL1 (Lys538) here Brain and muscle Arnt-like protein-1 (BMAL1) is a transcription factor known to regulate circadian rhythm. BMAL1 is acetylated at Lys538 upon heterodimerization with CLOCK to effect regulation of glucose homeostasis and metabolism. This BMAL1 antibody recognizes BMAL1 from multiple species in Western blots.

Acetyl-Histone H3 Immunoprecipitation (ChIP) Assay Kit Immunoprecipitate transcriptionally active chromatin from mammalian cells using anti-acetyl-Histone H3. Detection of the gene or promoter of interest in immunoprecipitated chromatin must be empirically determined using quantitative PCR or Southern blot analysis with promoter-specific primers or probe.

Anti-acetyl C/EBPβ (Lys215, Lys216) here The transcription factor CCAAT/Enhancer Binding Protein beta (C/EBPβ, also known as NFIL6) is a highly conserved B-Zip family member. C/EBPβ regulates multiple genes involved in hepatic function, immune function and adipogenesis, and acetylation at Lysines 215 and 216 may determine its specificity of action.

HDAC Assay Kit, fluorometric detection 384 well format This 384 well format kit is a newer version of the tried and true non-radiographic HDAC assay. Histone acetylation/deacetylation alters chromosome structure and affects transcription factor access to DNA. Routinely evaluated by deacetylation of HDAC Assay Substrate using HeLa nuclear extract.
