Background InformationZyxin, is a cytosolic, focal adhesion protein belonging to the Ajuba family and containing 3 LIM zinc-binding domains. It associates with the actin cytoskeleton near the adhesion plaques and, during mitosis, with microtubules. Zyxin localizes to stress fibers, cell-cell junctions and colocalizes with vinculin, paxillin and VASP at focal adhesions but not at the tips of lamellipodia. Zyxin is involved in the regulation of actin dynamics and is a likely component of the signal transduction pathway mediating adhesion-stimulated changes in gene expression. Zyxin displays an actin nucleating activity that is dependent on its amino terminal proline-rich motifs and independent of Arp2/3 complex. Zyxin appears to function as an adaptor or docking protein to organize protein complexes important for regulated actin cytoskeleton dynamics.

Zyxin is a zinc-binding phosphoprotein that concentrates at focal adhesions and along the actin cytoskeleton. Zyxin has an N-terminal proline-rich domain and three LIM domains in its C-terminal half. The proline-rich domain may interact with SH3 domains of proteins involved in signal transduction pathways while the LIM domains are likely involved in protein-protein binding. Zyxin may function as a messenger in the signal transduction pathway that mediates adhesion-stimulated changes in gene expression and may modulate the cytoskeletal organization of actin bundles.
