Background InformationVitamin D3 receptor (UniProt P11473; also known as 1,25-dihydroxyvitamin D3 receptor, Nuclear receptor subfamily 1 group I member 1, VDR) is encoded by the VDR (also known as NR1I1) gene (Gene ID 7421) in human. VDR is a member of the superfamily of nuclear receptors for steroid hormones and acts as a ligand-inducible transcription factor that targets vitamin D responsive elements. Upon ligand binding, VDR complexes with the retinoid-X receptor (RXR), the heterodimer then targets hormone-response elements, resulting in transcription or transrepression of specific genes. Ligand binding also induces a conformational change at the C-terminal ligand-binding domain (LBD; a.a. 192-427), resulting in recruitment of the p160/SRC family coactivators whose acetylase activity plays an important role in transcription modulations via chromatin remodeling. VDR gene mutations are associated with type II vitamin D-resistant rickets and alopecia (hair loss).
