Background InformationInterleukin-1 beta (UniProt; also known as IL-1 beta, IL-1b, IL-1beta) is encoded by the Il1b gene (Gene ID 16176) in murine species. Interleukin-1 (IL-1) is a cytokine that plays a central role in the regulation of acute and chronic inflammation. Two forms of IL-1, IL-1 and IL-1 , are transcribed from two separate genes and mediate similar, but not completely overlapping, biological effects via the membrane-bound IL-1 type 1 receptor (IL-1R1). Studies utilizing Il1a- and Il1b-knockout mice showed that IL-1 , but not IL-1 , is involved in T-cell-dependent antibody production via antigen-specific T-cell activation. IL-1 binding to the extracellular domain of IL1R1 promotes the recruitment of the IL-1 receptor accessory protein and subsequent signal transduction mediated by the cytoplasmic domains of IL-1R1 and IL1-R2. The IL-1 type 2 receptor (IL1R2) is unable to initiate signaling and only acts as a decoy receptor. Soluble receptors for IL-1 (sIL-1Rs) are produced by proteolytic cleavage of membrane-bound receptors or by alternative splicing and act as natural inhibitors by competing against membrane-bound IL1Rs for IL-1 binding. Murine IL-1 beta is produced with a propeptide sequence (a.a. 1-117), which is removed posttranslationally to yield the mature (a.a. 118-269) protein.
