Application NotesImmunohistochemistry (acetone fixation): Effective for human tissue, especially in circumstances where up-regulation of the receptor is being evaluated. The antibody does not stain formalin-fixed or paraffin-embedded tissues, and may not detect the low-level basal expression of alpavbeta6 in epithelia of healthy adults.

Immunocytochemistry: Effective for F6, HT-29, H441 cell lines (others not tested).

Flow cytometry: Effective for detection of human and monkey alphaVbeta6

Not effective for immunoprecipitation or western blot.

Optimal working dilutions must be determined by end user.

SpecificityMonoclonal antibody MAB2074Z, raised by immunizing Balb/c mice with recombinant secreted human avb6, is useful for flow cytometry and is especially useful for immunocytochemistry. The antibody works well for immunohistochemistry of human tissue, especially under circumstances where upregulation of the receptor is being evaluated. The antibody does not stain formalin-fixed or paraffin-embedded tissues, and may not detect the low-level basal expression of alphavbeta6 in epithelia of healthy adults. This antibody does not recognize the alphaV subunit and does not react with alphaV integrins other than alphavbeta6. Because the antibody is not effective in western blot, it is unknown whether the epitope recognized by the antibody localizes to the alphavbeta6 heterodimer or to the beta6 subunit only.

The I-domain containing integrin alpha V undergoes post-translational cleavage to yield disulfide-linked heavy and light chains, that combine with multiple integrin beta chains to form different integrins. Among the known associating beta chains (beta chains 1,3,5,6, and 8; 'ITGB1', 'ITGB3', 'ITGB5', 'ITGB6', and 'ITGB8'), each can interact with extracellular matrix ligands; the alpha V beta 3 integrin, perhaps the most studied of these, is referred to as the Vitronectin receptor (VNR). In addition to adhesion, many integrins are known to facilitate signal transduction.
