Background InformationInterleukin-12 subunit beta (UniProt: P29460; also known as IL-12B, Cytotoxic lymphocyte maturation factor 40 kDa subunit, CLMF p40, IL-12 subunit p40, NK cell stimulatory factor chain 2, NKSF2) is encoded by the IL12B (also known as NKSF2) gene (Gene ID: 3593) in human. IL-12 is a heterodimeric protein composed of the p35 and p40 subunits. It is secreted by activated monocytes, macrophages, and dendritic cells, and has been shown to target naïve, resting CD4+ T cells to promote their proliferation and secretion of cytokines. The IL-12 p40 subunit is a secreted protein, which is synthesized with a signal peptide that is subsequently cleaved off to generate the mature form. It can homodimerize and the homodimeric form is shown to be about 25 to 50-fold more active than the monomer in competing with native IL-12 in receptor binding. IL-12 p40 can act as a growth factor for activated T and NK cells and can enhance the lytic activity of NK/lymphokine-activated killer cells and stimulate the production of IFN-gamma by resting peripheral blood mononuclear cells. It is also reported to associates with IL23A to form the IL-23 interleukin that functions in innate and adaptive immunity. (Ref.: Stallmach, A., et al. (2004). Gut. 53(3): 339-345).
