Application NotesImmunohistochemistry: Suitable for staining frozen sections and paraffin wax embedded tissue. Protease treatment of paraffin embedded sections is required prior to antibody application. Frozen sections (7 µm) of placental tissue fixed with acetone for 1 hr. Antibody used at 1:10 dilution (Sato et al., 2003).

Immunocytochemistry: isolated EVT cells fixed with 0.5% PFA at 4°C for 15 minutes followed by acetone at -20°C for 2 minutes. Antibody was used at 1:10 dilution (Sato et al., 2003).

Flow cytometry: 10μL of conjugate per test.

Optimal working dilutions must be determined by the end user.

SpecificityThe antibody reacts with cytokeratin 7 which has been found in columnar and glandular epithelium of the lung, cervix, breast, bile ducts and larger collecting ducts of the kidney. Occasionally staining of blood vessel cell walls, particularly endothelial cells, may be observed.

The type II cytokeratins consist of basic or neutral proteins which are arranged in pairs of heterotypic keratin chains coexpressed during differentiation of simple and stratified epithelial tissues. This type II cytokeratin is specifically expressed in the simple epithelia lining the cavities of the internal organs and in the gland ducts and blood vessels. The genes encoding the type II cytokeratins are clustered in a region of chromosome 12q12-q13. Alternative splicing may result in several transcript variants; however, not all variants have been fully described.
