      Overview

      Replacement Information
      Description
OverviewNative vitronectin purified from human plasma. Extracellular matrix protein that promotes cell adhesion. Exists as a mixture of 75 kDa and 65 kDa polypeptides. Biochemically and immunologically distinct from fibronectin. Binds to cells through the RGD sequence in its cell-binding domain.
Intact protein and SDS-PAGE bands react with monoclonal antibodies to vitronectin, bind to heparin, and promote cell adhesion.
      M.W. 65,000 and 75,000.

      Recommended reaction conditions
Coating Tissue Culture Dishes with Vitronectin Although the optimal concentration for cell attachment and culture may differ for various cell types and for individual cell culture systems, typically concentrations as low as 50 ng/cm2 promote cell attachment and growth. Coating Procedure 1. Dissolve the desired amount of vitronectin in serum-free medium or PBS. The volume should be sufficient to coat the desired surface evenly (e.g. 1 ml for a 35 mm dish, 3 ml for a 60 mm dish). 2. Add the appropriate amount of distilled material to the culture surface. 3. Incubate at room temperature for 1-2 h. 4. Aspirate the remaining media. 5. Rinse the plates carefully. Avoid scratching the bottom surface of the plates. Store in the refrigerator (4°C) damp or air dried under sterile conditions. This protocol is meant to serve only as a guideline and may need to be modified for specific applications.
