      OverviewHuman TWEAK is a new secreted ligand in the tumor necrosis factor family that weakly induces apoptosis. TWEAK, like many members of the TNF family, was able to induce interleukin-8 synthesis in a number of cell lines. GF102 is a 17.0 kDa protein, comprising the TNF-like extracellular domain of TWEAK.

      Specific ActivityBIOLOGICAL ACTIVITY: <br /><br />GF102 is fully biologically active when compared to standards. The ED50 as determined by the dose-dependent stimulation of IL-8 production by Human PBMC is less than 10ng/ml. <br /><br />GF102 weakly induces the death of HT29 cells when cultured in the presence of IFN-g. The ED50 for this effect is between 30-45 ng/ml.

This cytokine has overlapping signaling functions with TNF, but displays a much wider tissue distribution. This cytokine can induce apoptosis via multiple pathways of cell death in a cell type-specific manner. This cytokine is also found to promote proliferation and migration of endothelial cells, and thus acts as a regulator of angiogenesis.

      Storage ConditionsThe lyophilized protein is stable for a few weeks at room temperature, but best stored at -20°C. Freeze/thaw cycles will result in significant loss of activity. After a quick spin, reconstitute in 10 mM NaP pH 7.5 to a concentration of 1 mg/ml. This solution can be diluted into other buffer solutions or stored at -20°C. Reconstituted TWEAK should be stored at -20°C or lyophilized in working aliquots.
