      Application CommentsDoes not recognize unphosphorylated RhoA. The immunogen sequence is conserved in rat and mouse RhoA, but cross-reactivity has not been tested. The immunogen sequence is not found in the RhoB and RhoC sequences. For immunoblotting, the membrane was incubated with antibody diluted in PBS/5% nonfat milk/0.04% Tween® 20 detergent for 1 h at room temperature. Antibody should be titrated for optimal results in individual systems.

      DescriptionImmunoaffinity purified rabbit polyclonal antibody. Recognizes the ~22 kDa RhoA protein when expressed as a GST fusion protein (total ~49 kDa) and phosphorylated in vitro by PKA.
      BackgroundRho (A, B, and C) proteins are members of the Ras superfamily of GTPases. They regulate a variety of cellular functions, including cell cycle progression, cytoskeletal rearrangement, and gene expression. Rho cycles between the active GTP-bound form and the inactive GDP-bound form. Conversion from the inactive to the active form and vice versa is controlled by guanine nucleotide exchange factors (GEFs) and GTPase-activating proteins (GAPs). The three Rho proteins are highly homologous and contain the consensus sequences necessary for GDP/GTP binding and GTPase activity. RhoA is phosphorylated at Ser188 in vitro by PKA and PKG. This phosphorylation enhances RhoGDI binding and inhibits RhoA-mediated stress fiber formation, indicating that Ser188 is an important site for negative regulation of RhoA activity.
