      Background InformationUromodulin (UniProt: P07911; also known as Tamm-Horsfall urinary glycoprotein, THP) is encoded by the UMOD gene (Gene ID: 7369) in human. Uromodulin is a homodimeric glycoprotein that is produced in the kidney and can polymerize into long filaments. It is expressed in the tubular cells of the kidney and is most abundant protein in normal urine. It plays a role in biogenesis and organization of the apical membrane of epithelial cells of the thick ascending limb of Henle's loop, where it promotes formation of complex filamentous gel-like structure that may play a role in the water barrier permeability. It also facilitates neutrophil migration across renal epithelia. Uromodulin contains a ZP domain (aa 334-589) that mediates polymerization, leading to the formation of long filaments. The core of the filament consists of stacked ZP domains, which assemble into a helical structure. Each ZP domain consists of an N-terminal (ZP-N; aa 334-429) and C-terminal (ZP-C; aa 465-589) region connected by a flexible linker. The linker allows the ZP domain to wrap around the ZP-C subdomain of the preceding subunit. It is proteolytically cleaved at a conserved C-terminal proteolytic cleavage site to generate the secreted form found in the urine. In the urine, it may contribute to colloid osmotic pressure, retard passage of positively charged electrolytes, and prevent urinary tract infection. It also inhibits the formation of liquid containing supersaturated salts and subsequent formation of salt crystals.
