      Background InformationHistone-lysine N-methyltransferase PRDM16 (UniProt: Q9HAZ2; also known as EC:2.1.1.367, PR domain zinc finger protein 16, PR domain-containing protein 16, Transcription factor MEL1, MDS1/EVI1-like gene 1) is encoded by the PRDM16 (also known as KIAA 1675, MEL1, PFM13) gene (Gene ID: 63976) in human. PDRM16 is a transcription factor with 10 CeH2-type zinc finger domains. It is highly expressed in adipocytes where it is involved in browning and thermogenesis of adipocytes, the beigeing of adipocytes, the adipogenic differentiation of myoblasts, and the conversion of visceral adipocytes to subcutaneous adipocytes. Its expression is also observed in uterus, kidney, cardiomyocytes, and interstitial cells. It is reported to control a bidirectional cell fate switch between skeletal myoblasts and brown fat cells. Loss of PRDM16 from brown fat precursors is reported to cause a loss of brown fat characteristics and promote muscle differentiation. Conversely, ectopic expression of PRDM16 in myoblasts is shown to induce their differentiation into brown fat cells. It stimulates brown adipogenesis by binding to PPAR- and activating its transcriptional function. PDRM16 also displays histone methyltransferase activity and monomethylates lysine 9 of histone H3 (H3K9me1) in the cytoplasm. It is also known to function as a repressor of TGF-β signaling. Four isoforms of PDRM16 have been described that are produced by alternative promoter usage and alternative splicing.
