      DescriptionPurified mouse monoclonal antibody generated by immunizing BALB/c mice with the specified immunogen and fusing splenocytes with SP2/O-NS1 plasmacytoma cells. Recognizes the ~43 kDa latent and the ~31 kDa active forms of cathepsin B.
      BackgroundCathepsin B (CB) is a normal lysosomal proteinase which is expressed in all cells. It is a cysteine endoproteinase and is structurally and functionally related to the papain family of proteinases. CB is synthesized as an inactive 43 kDa pro-enzyme which, by removal of a 623 amino acid propeptide, is activated to the single-chain form of 31 kDa or the two-chain form of 25 and 5 kDa subunits. Mature CB is localized in the lysosomes where it functions in protein turnover but has also been shown to be capable of degrading extracellular matrix proteins at acidic and neutral pH. Endogenous inhibitors of cysteine proteinases, the cystatins and stefins, may play a major role in regulating proteinase activity. In tumor cells and in cells exposed to mitogens, CB displays altered cellular trafficking resulting in the secretion of the 43 kDa precursor. Elevated levels of CB correlate with malignancy, suggesting that this enzyme may be a useful prognostic marker for several types of human cancers.
