      DescriptionPurified mouse monoclonal antibody. Recognizes the ~56-58 kDa latent and the ~18 kDa active form of caspase-8.
      BackgroundCaspase-8 (ICE-LAP5, MACH, FLICE, CAP4) is the most proximal of the members of the apoptotic caspase cascade. Caspase-8 is directly involved in apoptosis mediated through either Fas or the TNF receptor and forms one of the key components of the transducing DISC, the CAP4 protein. Cell lines that are deficient in caspase-8 are resistant to Fas mediated apoptosis. Nine different isoforms of caspase 8 have been identified but the "a" and "b" isoforms show the highest expression at the level of protein in multiple tissues. Caspase-8 has a pH optimum of 6.8 and has a kcat/Km of 8.7 X 105 M-1 s-1. The pro-form of caspase-8 has a molecular weight of approximately 55 kDa and undergoes several processing steps to generate the active heterodimer of p18 and p10/12. In addition to activation via Fas, oligomerization of caspase-8 at the cell membrane and interaction with Granzyme B and caspase 10 will induce auto-processing and activation of the apoptotic pathway. The mouse caspase-8 gene has also been identified and is expressed in the adult as well as at high levels in the day 9.5 and day 17.5 embryo suggesting a role in development.
