      Application CommentsImmunoblotting using a human placental proteasome preparation yields two bands at ~29 kDa and ~32 kDa. In ELISA the antibody reacts with the peptide sequence TVWSPQGRLHQVEYAMEA encompassing the prosbox I motif common to α-type subunits. Antibody should be titrated for optimal results in individual systems.

      DescriptionPurified mouse monoclonal antibody generated by immunizing BALB/c mice with the specified immunogen and fusing splenocytes with mouse myleloma cells. Recognizes the ~29-32 kDa α1, 2, 3, 5, 6 and 7 subunits of the 20S proteasome.
      BackgroundProteasomes are large multi-subunit complexes, that are localized in the nucleus and cytosol, and selectively degrade intracellular proteins. They play a major role in the degradation of many proteins involved in cellular processes such as cell cycling, apoptosis, and proliferation. The human proteasome contains two copies of each of seven different α-type and β-type subunits assembled to form a stack of four seven-membered rings. The α-subunits comprise the outer rings while the β-subunits comprise the inner rings. The α-subunits contain information for the binding of regulatory complexes such as PA700 and PA28.
