DescriptionAnti-phospho-Axin-1 (Thr485) Antibody
Alternate Names
  • Axin-1
  • Axis inhibition protein 1
  • Protein Fused
Background InformationAxin-1 (UniProt P57094; also known as Axis inhibition protein 1, Protein Fused) is encoded by the Axin1 (also known as Axin, Fu) gene (Gene ID 12005) in murine species. The expansion of the mammalian cerebral cortex is controlled by a balance between the proliferation and differentiation of intermediate progenitors (IPs). Axin-1 is a Wnt signaling pathway scaffold protein that plays a critical role in orchestrating the signals controlling IP population and the number of differentiated neurons during neurogenesis in the developing cerebral cortex. Axin-1 is phosphorylated at Thr485 by Cdk5, the Thr485 phosphorylation results in Axin-1 translocation from the cytoplasm to nucleus upon neuronal progenitor cell (NPC) differentiation. The interaction between cytoplasmic Axin-1 and GSK-3β is critical for the amplification of the undifferentiated IP pool, whereas the interaction between phosphorylated Axin-1 and β-catenin in the nucleus promotes neuronal differentiation. Cdk5-mediated Axin-1 phosphorylation therefore serves as a molecular switch from amplification to differentiation in NPCs.

Applications
ApplicationDetect phospho-Axin-1 using this Anti-phospho-Axin-1 (Thr485) Antibody validated for use in Western Blotting, Immunofluorescence.
Key Applications
  • Western Blotting
  • Immunofluorescence
Application NotesImmunofluorescence Analysis: An 1:500 from a representative lot detected nucleus localized axin-1 phosphorylation (pAxin) in paraformaldehyde-fixed, O.C.T-embedded adult mouse brain cryosections by fluorescent immunohistochemistry (Courtesy of Professor Nancy Y. Ip, Hong Kong University of Science and Technology).
Western Blotting Analysis: An 1:1,000 from a representative lot detected a develpmental age-dependent axin-1 phosphorylation level in total brain lysates from E18 mouse embryo, postnatal P10 and adult mice (Courtesy of Professor Nancy Y. Ip, Hong Kong University of Science and Technology).
Immunofluorescence Analysis: A representative lot detected phosphorylated axin-1 (pAxin) in the nuclei of intermediate progenitors (IPs) in the developing cortex by fluorescent immunohistochemistry using paraformaldehyde-fixed, O.C.T-embedded cryosections. The proportion of pAxin-positive neural progenitor cells (NPCs) increased progressively from E13.5 to E15.5 (Fang, W.Q., et al. (2013). Neuron 79(4):665–679).
Western Blotting Analysis: A representative lot detected phosphorylated axin-1 in the nuclear, but not cytosolic, fraction of E13.5 mouse embryo brain and cultured neural progenitor cells (NPCs). A significant upregulation of nuclear phosphorylated axin-1 was observed in NPCs upon differentiaion induction in differentiation medium (DM) than in undifferentiated/proliferating NPCs in proliferation medium (PM) (Fang, W.Q., et al. (2013). Neuron 79(4):665–679).
Western Blotting Analysis: A representative lot detected the phosphorylation of murine wild-type axin-1, but not T485A mutated axin-1, by CDK5/p25 in immune-complex kinase assays (Fang, W.Q., et al. (2011). J. Neurosci. 31(38):13613–13624).
Western Blotting Analysis: A representative lot detected a greatly diminished axin-1 phosphorylation in mouse hippocampal neurons treated with the Cdk inhibitor Roscovitine (Cat. No. 557360), as well as in neurons from Cdk5-/- or Cdk5 activator p35-/- knockout mice (Fang, W.Q., et al. (2011). J. Neurosci. 31(38):13613–13624).
Western Blotting Analysis: A representative lot detected an induction of axin-1 phosphorylation in serum-starved mouse hippocampal neurons following brain-derived neurotrophic factor (BDNF) or neurotrophin-3 (NT-3) treatment (Fang, W.Q., et al. (2011). J. Neurosci. 31(38):13613–13624).

HostRabbit
SpecificityThis polyclonal antibody detected CDK5/p25-dependent axin-1 Thr485 phosphorylation both in vitro and in vivo (Fang, W.Q., et al. (2011). J. Neurosci. 31(38):13613–13624). The Thr485 designation was originally assigned based on mouse (NP_001153070.1) and rat (NP_077381.1) sequences deposited in NCBI protein database. The numbering can vary depending on species and the source of published sequence. For example, the target resiude corresponds to Thr481 of the human sequence and Thr480 of the mouse and rat sequences reported by UniProt (O15169, O35625 and O70239, respectively).
