Background InformationLysine-specific demethylase 6A (UniProt O15550; also known as Histone demethylase UTX, Ubiquitously transcribed tetratricopeptide repeat protein X-linked, Ubiquitously-transcribed TPR gene on the X chromosome, Ubiquitously transcribed X chromosome tetratricopeptide repeat protein) is encoded by the KDM6A (also known as bA386N14.2, KABUK2, UTX) gene (Gene ID 7403) in human. Histone lysine demethylases are divided into the flavin adenine dinucleotide-/FAD-dependent amine oxidases that target mono-, di-, but not tri-, methylated histone lysine residues and the Jumonji C/JmjC domain-containing enzymes that target specific mono-, di- and tri-methylated histone lysine residues. The JmjC domain enzymes rely on α-ketoglutarate (α-KG), Fe(II), and molecular oxygen as cofactors in the demethylation reaction. KDM6A/UTX, KDM6B/JMJD3, and UTY constitute the KDM6 subfamily of JmjC domain-containing histome demethylases. KDM6A/UTX and KDM6B/JMJD3 target H3K27me3 and H3K27me2, whereas UTY display little catalytic activity.
