Background InformationExtracellular sulfatase Sulf-2 (UniProt: Q8IWU5; also known as hSulf-2, Arylsulfatase, EC:3.1.6.13, N-acetylglucosamine-6-sulfatase) is encoded by the SULF2 (also known as KIAA1247 and UNQ559/PRO1120) gene (Gene ID: 55959) in human. Sulfatase 1 (Sulf-1) and sulfatase 2 (Sulf-2) are heparan sulfatase editing sulfatases that remove 6-O sulfate moiety within heparan sulfate (HS) chains and alter the affinity of the HS chains for growth factors and growth factor receptors. Sulf-2 is synthesized with a signal peptide (aa 1-24) that is subsequently cleaved off to produce the mature protein. Its N-terminal region contains the catalytic domain (aa 1-415) that is followed by a hydrophilic domain (aa 416-715) and the C-terminal region. The hydrophilic domain is necessary for its endoglucosamine-6-sulfatase activity. Glycosylation at serine 583 is shown to negatively regulate its N-acetylglucosamine-6-sulfatase and arylsulfatase activities. Higher expression of Sulf-2 is reported in the ovary, skeletal muscle, stomach, brain, uterus, heart, kidney, and placenta. It is detected in the endoplasmic reticulum, Golgi apparatus, and on the cell surface. A secreted form has also been described that is produced after processing by furin. In a majority of hepatocellular carcinomas (HCC) Sulf-2 enhances tumor growth and progression via activation of the Wnt and TGF-β signaling. It has broad oncogenic effects in HCC, and it increase cell growth by upregulating glypican 3 that promotes release of FGF2 and Wnt form glypican 3 to bind to their cognate receptors and enhance receptor tyrosine kinase activity and Wnt/β-catenin signaling. Knockout of Sulf-2 is reported to suppress HCC tumorigenesis. Two isoforms of Sulf-2 have been described that are produced by alternative splicing. (Ref.: Luo, X., et al. (2021).
