Background InformationDeoxynucleoside triphosphate triphosphohydrolase SAMHD1 (UniProt Q9Y3Z3; also known as DCIP, Dendritic cell-derived IFNG-induced protein, dNTPase, Monocyte protein 5, MOP-5, SAM domain and HD domain-containing protein 1) is encoded by the SAMHD1 (also known as CHBL2, HDDC1, MOP5, SBBI88) gene (Gene ID 25939) in human. The sterile alpha motif domain and histidine-aspartate domain-containing protein 1 (SAMHD1) is a dGTP/GTP-activated deoxynucleotide (dNTP) triphosphohydrolase regulated by Cyclin A2/CDK1-dependent phosphorylation and is involved in balancing cellular dNTP pools. SAMHD1 was originally identified as a disease gene associated with the rare infantile encephalopathy Aicardi-Goutières syndrome. SAMHD1 was later shown to restrict HIV-1 replication in non-dividing myeloid-lineage cells and resting T cells by reducing the cellular dNTP concentration. Simian immunodeficiency viruses (SIVs) are found to use their Vpx/Vpr accessory proteins to engage and modify host Cullin-RING-type E3 ubiquitin ligases substrate recognition to target SAMHD1 for poly-ubiquitylation and subsequent proteasomal degradation.
