Background InformationRho-related GTP-binding protein RhoE (UniProt: P61588; also known as Rho family GTPase 3, Rnd3) is encoded by the Rnd3 (also known as Arhe, Rhoe) gene (Gene ID: 74194) in murine species. RhoE is a multi-functional protein that is a member of the Rho GTPase family and is involved in the control of actin cytoskeletal dynamics. It is reported to bind to but does not hydrolyze GTP. Hence, it is not regulated by the classical GTP/GDP conformational switch of small GTPases. It is widely expressed; however, its expression levels vary significantly between different cell types. RhoE is found in both membrane and cytosolic fractions and is also reported to be localized to the Golgi complex and plasma membrane. RhoE activity is largely regulated by phosphorylation. It is shown to inhibit RhoA signaling by binding to the kinase domain of ROCK1, thereby preventing it from phosphorylating its targets. RhoE can undergo phosphorylation on multiple sites by ROCK1, which leads to its enhanced stability. Phosphorylation of RhoE is shown to diminish its association with membranes. RhoE phosphorylated on serine 11 is localized exclusively in the cytosol, whereas unphosphorylated RhoE is present in cell membranes. (Ref.: Riento, K., et al. (2005). Biochem. Soc.
