Background InformationRAB35 is a member of the Rab GTPases identified in the human genome. It is ubiquitously expressed and regulates a fast endocytotic recycling events associated with the terminal steps of cytokinesis, the formation of immunological synapse, and peptide loading onto major histocompatibility complex III molecules. RAB35 also plays a significant role in the formation of the immunological synapse (IS) between peptide-loaded antigen presenting cells (APCs) and T cells.

UniProt SummaryFUNCTION: In the process of endocytosis, essential rate-limiting regulator of a fast recycling pathway back to the plasma membrane. During cytokinesis, required for the postfurrowing terminal steps, namely for intercellular bridge stability and abscission, possibly by controlling phosphatidylinositol 4,5-bis phosphate (PIP2) and SEPT2 localization at the intercellular bridge.

SUBUNIT STRUCTURE: Interacts with DENND1A and DENND1B in a nucleotide-dependent manner. The low binding to DENND1C is nucleotide-independent. All 3 DENND1 act as GEFs for RAB35 and thus activate the protein.

SUBCELLULAR LOCATION: Cell membrane; Lipid-anchor; Cytoplasmic side Potential. Membrane › clathrin-coated pit. Cytoplasmic vesicle › clathrin-coated vesicle. Endosome. Melanosome. Note: Present on sorting endosomes and endosomal tubules. Tends to be enriched in PIP2-positive cell membrane domains. During mitosis, associated with the plasma membrane and present at the ingressing furrow during early cytokinesis as well as at the intercellular bridge later during cytokinesis. Identified by mass spectrometry in melanosome fractions from stage I to stage IV.
