Application CommentsDoes not cross-react with recombinant human inducible NOS (iNOS) or neuronal NOS (nNOS). Variables associated with assay conditions will dictate the proper working dilution.

Recommended Procedure for Immunoblotting of Anti-eNOS:

1. Run SDS-PAGE using 4-15% gradient gels or single percentage gels (e.g., 7.5%) and electrophoretically transfer to PVDF membrane.

2. Block overnight with 4% normal goat serum in evaporated goat milk diluted 1:5 with TBS/Tween®-20 detergent buffer.

3. Wash 2X with TBS/Tween®-20 detergent.

4. Apply the reconstituted rabbit anti-eNOS at a dilution of at least 1:500 in the following diluent: 1% normal goat serum in evaporated goat milk diluted 1:5 with TBS/Tween®-20 detergent buffer. Higher dilutions of the primary antibody may be required. Incubate 2-4 h.

5. Wash 3X with TBS/Tween®-20 detergent.

6. Apply HRP-conjugated, affinity-purified goat anti-rabbit IgG at the recommended dilution. Use 1% normal goat serum in evaporated goat milk diluted 1:5 with TBS/Tween®-20 detergent as the diluent. Incubate 1-2 h.

7. Wash 3X with TBS/Tween®-20 detergent, then soak overnight in a reasonably large volume of the same buffer.

8. Develop the blot using an HRP substrate suitable for immunoblotting, such as DAB.
