Background InformationPlasmalemmal neurotransmitter transporters sequester synaptic and peri synaptic transmitter into presynaptic elements. The Dopamine Transporter (DAT) is responsible for the reaccumulation of dopamine after it has been released. Levels of DAT protein expression are altered by chronic drug administration. The activity of the DAT reuptake carrier is sodium dependent, and it is suspected to play a role in such neurologic and psychiatric disorders as Parkinson's disease, Tourette's disease, schizophrenia, and addiction. It is a 12 transmembrane domain transporter with the N and C terminal regions located within the cytoplasm.

Application NotesWestern blot: 1-10 μg/mL using ECL. An antibody raised against a similar peptide has been shown to react with a broad 75-80 kDa band in rat striatum (Hastrup et al., 2001) and not cerebellum. Rat DAT contains various carbohydrate moieties and neuraminidase and N-Glycanase treatment reduces its molecular weight to about 55 kDa.

Immunoprecipitation: 2-10 μg of antibody (Chang et al., 2001)

Immunohistochemistry: 1:250-1:500 on frozen sections of tissues fixed with 4% paraformaldehyde (Chang et al., 2001; Ourednik, et al., 2002). 0.05% trypsin for 10-15min has been found effective in exposing the epitope in some preparations.

ELISA: 0.5-1.0 μg/mL using 1 μg/mL of DAT control peptide (Catalog Number AG296) per well.

Optimal working dilutions must be determined by end user.
Biological Information
ImmunogenAn 18 amino acid peptide sequence near the NH2-terminus of rat brain dopamine transporter coupled to KLH (Kilty et al. 1991; Giros et al. 1991; Shimada et al. 1991; Kitayma 1992). This region is presumably the cytoplasmic domain.
