Description
Catalogue NumberABE374
DescriptionAnti-DAZ Antibody
Alternate Names
  • Deleted in azoospermia protein 1
Background InformationDAZ is an RNA-binding protein that belongs to the DAZ family of proteins required for normal growth, development, and fertility. The DAZ family of proteins have one or more RNA recognition motif (RRM) domains and multiple DAZ repeats. Mammalian DAZ is widely expressed in adults with high levels of expression in testis. Frequent deletion of the DAZ genes suggests that DAZ plays an important role in spermatogenesis. It was even proposed that DAZ family proteins are involved in mRNA metabolism during spermatogenesis.

Applications
ApplicationAnti-DAZ Antibody is a Rabbit Polyclonal Antibody for detection of DAZ also known as Deleted in azoospermia protein 1 & has been validated in WB, IHC(P).
Key Applications
  • Western Blotting
  • Immunohistochemistry (Paraffin)
Application NotesImmunohistochemistry Analysis: A 1:400 dilution from a representative lot detected DAZ in human testis tissue.
Biological Information
ImmunogenKLH-conjugated linear peptide corresponding to the N-terminus of human DAZ.
EpitopeN-terminus
ConcentrationPlease refer to the Certificate of Analysis for the lot-specific concentration.
HostRabbit
SpecificityThis antibody recognizes the N-terminus of DAZ. This antibody demonstrates cross-reactivity with DAZ2, DAZ3, DAZ4, and DAZL each of which contains repeats of the DAZ gene.

Entrez Gene Number
Entrez Gene SummaryThis gene is a member of the DAZ gene family and is a candidate for the human Y-chromosomal azoospermia factor (AZF). Its expression is restricted to premeiotic germ cells, particularly in spermatogonia. It encodes an RNA-binding protein that is important for spermatogenesis. Four copies of this gene are found on chromosome Y within palindromic duplications; one pair of genes is part of the P2 palindrome and the second pair is part of the P1 palindrome. Each gene contains a 2.4 kb repeat including a 72-bp exon, called the DAZ repeat; the number of DAZ repeats is variable and there are several variations in the sequence of the DAZ repeat. Each copy of the gene also contains a 10.8 kb region that may be amplified; this region includes five exons that encode an RNA recognition motif (RRM) domain. This gene contains three copies of the 10.8 kb repeat. However, no transcripts containing three copies of the RRM domain have been described; thus the RefSeq for this gene contains only two RRM domains.

UniProt Number
UniProt SummaryFUNCTION: RNA-binding protein that plays an essential role in spermatogenesis. May act by binding to the 3'-UTR of mRNAs and regulating their translation. Promotes germ-cell progression to meiosis and formation of haploid germ cells.

SUBUNIT STRUCTURE: Forms a heterodimer with BOLL and DAZL. Interacts with PUM2, DAZAP1, DAZAP2, DZIP1 and DZIP3.

SUBCELLULAR LOCATION: Cytoplasm. Nucleus. Note: Predominantly cytoplasmic. Nuclear at some stages of spermatozoide development. Localizes both to the nuclei and cytoplasm of spermatozoide differentiation. Nuclear in fetal gonocytes and in spermatogonial nuclei. It then relocates to the cytoplasm during male meiosis.

TISSUE SPECIFICITY: Testis-specific. Expression restricted to premeiotic germ cells, particularly in spermatogonia (at protein level).
Molecular Weight~83 kDa calculated. Additional bands due to polymorphic expression of DAZ as well as cross-reactivity with DAZ2, DAZ3, DAZ4, and DAZL with calculated molecular weights of ~33 kDa, 37 kDa, 75 kDa, and 100 kDa may be observed in some cell lysates (Poongothai, J., et al. (2009). Singapore Med J. 50(4):336-347; Kim, B., et al. (2009). Hum Reprod. 24(6):1507-1515). This antibody detects DAZ proteins at ~36 kDa, ~80 kDa, and ~105 kDa.
