Alternate Names
  • BSPII
  • Integrin-binding Sialoprotein
Background InformationBone Sialoprotein II is one of the major noncollagenous proteins in the extracellular matrix of bone. It is a phosphorylated glycoprotein with an approximate molecular weight of 70 kDa. The protein has been found in osteoblasts and osteocytes. Diseases concerning bone turnover, pathological bone alterations, as well as the rapid increase of osteoporosis make it necessary to establish new markers. Bone Sialoprotein is discussed as a potential serum marker for monitoring bone remodeling.

Entrez Gene Number
Entrez Gene SummaryThe protein encoded by this gene is a major structural protein of the bone matrix. It constitutes approximately 12% of the noncollagenous proteins in human bone and is synthesized by skeletal-associated cell types, including hypertrophic chondrocytes, osteoblasts, osteocytes, and osteoclasts. The only extraskeletal site of its synthesis is the trophoblast. This protein binds to calcium and hydroxyapatite via its acidic amino acid clusters, and mediates cell attachment through an RGD sequence that recognizes the vitronectin receptor.

UniProt Number
UniProt SummaryFUNCTION: SwissProt: P21815 # Binds tightly to hydroxyapatite. Appears to form an integral part of the mineralized matrix. Probably important to cell-matrix interaction. Promotes Arg-Gly-Asp-dependent cell attachment.
SIZE: 317 amino acids; 35018 Da
SUBCELLULAR LOCATION: Secreted.
PTM: N-glycosylated; glycans consist of sialated and core- fucosylated bi-, tri- and tetraantennary chains. & O-glycosylated at eight sites; mucin-type glycans contain Gal, GlcNAc, GalNAc and terminal NeuAc.
MISCELLANEOUS: It is possible that the segments of clustered carboxyl groups mediate the strong binding to hydroxyapatite.
