Novel non-ionic detergent useful for plasmid DNA isolation. Plasmid DNA isolated using APO-10 does not require the use of phenol/chloroform or proprietary resins. Provides excellent yields of high quality DNA. Allows plasmid DNA to be sequenced through difficult regions without the need to subclone into single-stranded vectors.

Recommended reaction conditions
Protocol for Plasmid DNA Isolation using APO-10 1. Centrifuge 1.5 ml of bacterial culture at 12,000 x g for 2 min. 2. Resuspend the pellet in 100 µl of 50 mM glucose, 25 mM Tris-HCl, 10 mM EDTA, 10 µg/ml RNase, pH 8.0. 3. Add 200 µl of 200 mM NaOH and 1% APO-10. Mix gently by inverting. Incubate at room temperature for 5 min. 4. Add 150 µl of 3 M potassium acetate buffer, pH 4.8. Mix gently by inverting. Incubate at room temperature for 5 min. 5. Centrifuge sample for 15 min at 4°C. Collect the supernatant. 6. Add 1 ml of 100% ethanol to the supernatant, mix, and centrifuge for 15 min at 4°C. Wash the pellet in 70% ethanol and dry. 7. The DNA pellet can be resuspended in 25 µl TE buffer (10 mM Tris-HCl, pH 8.0, 1 mM EDTA). RNase A can be added to digest any RNA.
