Read This Before You Start Your Extracellular Vesicle Research!

EV preparations can vary in molecular composition and function, depending on:
  • Cell culture viability
  • Apoptotic index
  • Cell proliferation rate
  • Status of the cell culture medium
  • Handling conditions
  • Other experimental parameters
To support EV researchers trying to navigate this rapidly changing field, we've made available to you a suite of resources for your easy reference.
  1. Wall Poster: Preparing functional extracellular vesicles (EVs): Join the rEVolution.

Hang this unique poster in your lab to be reminded of EV biogenesis pathways, cell type-specific EV processes, and characteristic protein and miRNA markers.

  2. Recorded Presentation from the American Association for Cancer Research (AACR) 2015 Annual Meeting: Strategies for enrichment, quantitation, and characterization of microvesicles for cancer research

In this 45-minute presentation, learn best practices for standardizing and reducing the EV preparation workflow, and eliminating contamination of EV preparations with dead-cell-derived vesicles. We will show how imaging flow cytometry improves the analysis and characterization of these microvesicles in a quantitative and reproducible manner above and beyond the current methods of analysis.

  3. Review article in BioProcessing Journal (in press): Preparing biotherapeutic extracellular vesicles: ultrafilters, MSCs and the regulatory horizon.

  4. Detection of internalized exosomes in single cancer cells using Amnis® imaging flow cytometry: View Poster Presented at CYTO 2013

Exosomes were co-cultured with tumor cells and analyzed for internalization of PKH26+ particles in the tumor cell lines using the Amnis ImageStream®X imaging flow cytometer.

ISEV Guidances: Quick Links

The International Society for Extracellular Vesicles (ISEV) has published guidances for preparation and analysis, recognizing that the exact methods could depend on the scientific question being asked.
