Description
A concentrate of dithiothreitol (DTT) in phosphate buffer. Useful for the isolation of pathogenic or saprophytic bacteria, fungi, yeast, and epithelial cells from sputum and other mucoid fluids.
Form
Liquid
Formulation
20 vials each containing 10 ml of concentrated DTT OR 1 vial containing 10 ml of concentrated DTT.
Recommended reaction conditions
A. To prepare sputum samples:
1. In a screw-cap centrifuge tube, overlay the sputum sample with an equal volume of the diluted SPUTOLYSIN® Reagent.
2. Suspend the sputum by mixing on a vortex mixer for 30 s.
3. Allow the suspended mixture to stand at room temperature for 15 min.
4. Centrifuge the mixture for 5 min at 1500 rpm. Discard the supernatant.
B. To prepare cultures for acid-fast bacilli:
1. Suspend the pellet from Step A(4) in 5 to 10 ml of 1% sodium hydroxide by mixing on a vortex mixer for 1 min.
2. Centrifuge the mixture for 15 min at 3000 rpm. Discard the supernatant.
3. Rinse the pellet twice in 10 ml of sterile normal saline to remove the residual sodium hydroxide.
4. Resuspend the pellet in 0.5 ml of the diluted SPUTOLYSIN® Reagent.
