      Background InformationProtein Themis (UniProt: Q8BGW0; also known as Grb2-associating protein, Gasp, Protein Thylex, Thymocyte-expressed molecule involved in selection) is encoded by the Themis gene (Gene ID: 210757) in murine species. T cell development in the thymus is shown to follow an ordered progression from CD4- 8- (double negative, DN) through CD4+8+ (double positive, DP) to single positive (SP, CD4+8 or CD8+4 ) stages, which depends on a complex interplay of signaling pathways. Themis plays an important role in late thymocyte development by controlling both positive and negative T-cell selection. It is required to sustain and/or integrate signals required for proper lineage commitment and maturation of T-cells. Themis is expressed in the thymus and to a lesser extent in the spleen but is not detectable in non-lymphoid tissues. It is highly expressed in thymocytes between the pre-T-cell antigen receptor (pre-TCR) and positive-selection checkpoints and expressed at low level in mature T-cells. Mice lacking Themis display a defect in the selection process mediated by the positive selection checkpoints, resulting in reduced numbers of SP cells and mature peripheral T cells. Themis contains two novel cysteine-based CABIT (cysteine-containing, all beta in Themis) domains (aa 1-260 and 261-520), a bipartite type nuclear localization sequence and a proline-rich sequence. CABIT domain modulate Themis by interacting directly with the PTP domain of SHP-1 and stabilize the catalytic cysteine residue of SHP-1, leading to inhibition of its tyrosine phosphatase activity.       UniProt SummaryFUNCTION: Plays a central role in late thymocyte development by controlling both positive and negative T-cell selection. Regulates T-cell development through T-cell antigen receptor (TCR) signaling and in particular through the regulation of calcium influx and phosphorylation of Erk.

      SUBUNIT STRUCTURE: Interacts with PLCG1, ITK and GRB2.

      SUBCELLULAR LOCATION: Cytoplasm. Nucleus

      TISSUE SPECIFICTY: Expressed in the thymus and to a lesser extent in the spleen but not detectable in nonlymphoid tissues. Highly expressed in thymocytes between the pre-T-cell antigen receptor (pre-TCR) and positive-selection checkpoints and expressed at low level in mature T-cells (at protein level).

      INDUCTION: Down-regulated by stimulation through the alpha-beta TCR.

      PTM: Phosphorylated on Tyr residues quickly after TCR stimulation.

      DISRUPTION: phenotype Mice are viable, are born at the expected mendelian frequency and present no gross abnormalities. They however display a blockade in late T-cell development with defects in thymocyte selection. The number of transitional CD4+CD8(int) thymocytes as well as CD4+ or CD8+ single-positive thymocytes is lower. Thymocytes show defective positive selection, resulting in fewer mature thymocytes. Negative selection is also impaired. A greater percentage of T-cells have CD4+CD25+Foxp3+ regulatory and CD62L(lo)CD44(hi) memory phenotypes compared to wild-type T-cells.
