      Background InformationATP-sensitive inward rectifier potassium channel 10 (UniProt P78508; also known as ATP-dependent inwardly rectifying potassium channel Kir4.1, Inward rectifier K(+) channel Kir1.2, Potassium channel, inwardly rectifying subfamily J member 10) is encoded by the KCNJ10 (also known as SESAMES) gene (Gene ID 3766) in human. Inward rectifier K+ channels (Kirs) modulate cell membrane potential and are classified as K+ transport channels (Kir1.x, Kir4.x, Kir5.x, Kir7.x), classic Kir (Kir2.x), G-protein-gated Kirs, and ATP-sensitive Kirs. Kirs mediate diverse physiological functions of both excitable and nonexcitable epithelial cells, such as cell adhesion-migration, volume regulation, apoptosis, and proliferation. Kir4.1 is widely expressed in the kidney distal convoluted tubules and neural glia cells, its expression in the cochlea and retina is also reported. Kir4.1 modulates plasma K+ by favoring K+ exit, and inhibition of Kir4.1 results in membrane depolarization. KCNJ10 gene mutations are known causes of SeSAMES (seizures, sensorineural deafness, ataxia, mental retardation, and electrolyte imbalance syndrome). Human Kir4.1 is a 379-amino acid multi-pass membrane (a.a. 65-89, 115-126, 143-164) protein containg two small extraceullar regions (a.a. 90-114 and 134-142), a pore-forming domain (a.a. 134-142), and having both N- and C-terminal exposed intracellularly (a.a. 1-64 and a.a. 165-379).
