Background InformationPigment epithelium-derived factor (PEDF) was first identified as a 50 kDa secreted protein in conditioned medium from cultured fetal human retinal pigment epithelium (RPE) cells (Tombran-Tink, 1991). More recently, PEDF was shown to have neurotrophic activity in the retina in vivo (Cayouette, 1999). PEDF is also a neurotrophic and neuroprotective factor in other systems, including cultured cerebellar granule cells, primary hippocampal neurons, spinal cord motor neurons and amphibian photoreceptors cultured in the absence of RPE cells (Karakousis, 2001). The binding of PEDF to retinoblastoma and cerebellar granule cells may be receptor-mediated. Recently, PEDF was shown to be a potent inhibitor of angiogenesis in endothelial cell cultures and in rat cornea and retina in vivo (Dawson, 1999; Stellmach, 2001).

      • ELISA
      • Immunocytochemistry
      • Immunohistochemistry
      • Western Blotting
      Application NotesWestern Blot:
Recognizes a protein with a molecular weight of ~46 kDa when tested against immunogen. Western blot positive control is available (Catalog number AG672)

      ELISA:
A previous lot of this antibody was used on ELISA.

      Immunocytochemistry:
A previous lot of this antibody was used on Immunocytochemistry.

      Immunohistochemistry:
A previous lot of this antibody was used on frozen sections.

Optimal working dilutions must be determined by end user.
      Biological Information
ImmunogenRecombinant human GST-PEDF fusion protein. The sequence corresponding to amino
acids 196-347 of human PEDF tagged with GST on the N-terminal was expressed in E. Coli. GST-PEDF is about 46 kDa on SDS-PAGE.

      UniProt Number
UniProt SummaryFUNCTION: SwissProt: P36955 # Neurotrophic protein; induces extensive neuronal differentiation in retinoblastoma cells. Potent inhibitor of angiogenesis. As it does not undergo the S (stressed) to R (relaxed) conformational transition characteristic of active serpins, it exhibits no serine protease inhibitory activity.
SIZE: 418 amino acids; 46342 Da
SUBCELLULAR LOCATION: Secreted. Melanosome. Note=Enriched in stage I melanosomes.
TISSUE SPECIFICITY: Retinal pigment epithelial cells and blood plasma.
DEVELOPMENTAL STAGE: Expressed in quiescent cells.
DOMAIN: SwissProt: P36955 The N-terminal (AA 44-121) exhibits neurite outgrowth- inducing activity. The C-terminal exposed loop (AA 382-418) is essential for serpin activity.
      PTM: The N-terminus is blocked.
SIMILARITY: Belongs to the serpin family
