Detection of KDEL by immunoblotting. Samples: Recombinant Grp78 (BiP) protein (lane 1), whole cell lysate RK-13 cells (lane 2), mouse liver microsomes (lane 3), rat liver microsomes (lane 4), and whole cell lysate from heat shocked HeLa cells (lane 5). Primary antibody: Anti-KDEL Mouse mAb (10C3) (Cat. No. 420400) (1:1000). Detection: chemiluminescence.
Description
Protein G purified mouse monoclonal antibody. Recognizes the KDEL Grp78 (BiP) and Grp94 proteins.
Background
Several resident proteins in the ER are involved in the proper assembly and folding of newly synthesized proteins. These include the glucose-regulated protein 94 (Grp94), BiP, which functions in protein oligomerization, and protein disulfide isomerase (PDI), which catalyzes the rearrangement of the disulfide bond. Grp94, BiP, and PDI share a carboxy-terminal sequence (Lys-Asp-Glu-Leu) with other resident ER proteins. The KDEL sequence is sufficient for the retention of a protein in the ER.
