      Background InformationEstrogen receptor (UniProt: P06211; also known as ER, ER-alpha, Estradiol receptor, Nuclear receptor subfamily 3 group A member 1) is encoded by the Esr1 (also known as Esr, Estr, Nr3a1) gene (Gene ID: 24890) in rat. Estrogen receptor a (ERa) is a steroid hormone receptor that is involved in the regulation of eukaryotic gene expression and affects cellular proliferation and differentiation in target tissues. Its expression has been reported in mammary gland, uterus, ovary, bone, prostrate, testes, and adipose tissue. In the absence of ligand, it is weakly associated with nuclear components and upon hormone binding its affinity is greatly enhanced. Its DNA binding region is localized to amino acids 190-255 (Domain C), which is highly conserved and contains two Cys2Cys2 zinc finger motifs. This region is essential and sufficient for binding of ER to the estrogen response element (ERE). Domain E, the ligand-binding domain (LBD; aa 316-552), is also highly conserved and is responsible for interaction of the receptor with various estrogens and antiestrogens. In ERa, transcription activation function-1 (AF-1) is localized in the amino-terminal A/B domain (aa 1-189) of the receptor and serves as a ligand-independent transcription activation domain. Activation function-2 (AF-2) resides in the hormone-binding domain (aa 316-600) and acts as a ligand-inducible activation domain. ERa can undergo phosphorylation by CDK2/cyclin A and by CK1 that enhances its transcriptional activity. In many breast cancers, ER activation by estrogens is shown to be responsible for enhanced proliferation, which is counteracted by the presence of ERβ that exerts an antiproliferative effect. (Ref.
