      Background InformationCell cycle and apoptosis regulator protein 2 (UniProt Q8N163; also known as Cell division cycle and apoptosis regulator protein 2, DBC-1, DBC.1, DBIRD complex subunit KIAA1967, Deleted in breast cancer 1, p30 DBC, p30 DBC protein) is encoded by the CCAR2 (also known as DBC1, KIAA1967, NET35) gene (Gene ID 57805) in human. Deleted in breast cancer gene 1 protein (DBC1) is a nuclear protein involved in regulating functions of nuclear receptors (NRs), including androgen receptor (AR) and estrogen receptor (ER) alpha and beta. DBC1 stabilizes the interaction between chicken ovalbumin upstream promoter transcription factor I (COUP-TFI) and NCoR by interacting directly with both proteins. DBC1 also interacts with NR complex interacting factor (NRC)-1 to drive the transcription regulation of NRs. Caspase-dependent processing of DBC1 promotes apoptosis, and depletion of DBC1 negatively regulates p53-dependent apoptosis through its specific inhibition of SIRT1. The N-terimus of DBC1 is shown to interact with BRCA1 C-terminal BRCT domain and the expression of DBC1 represses BRCA1-mediated transactivation of the SIRT1 promoter. DBC1 is also a negative regulator against the ligand-dependent transcriptional activation function of ERbeta. The DBC1 protein encoded by the CCAR2 gene is not the same as the protein encoded by the BRINP1 gene, which also goes by the same acronym DBC1 for "Deleted in bladder cancer protein 1".
