      DescriptionAnti-VSV-Glycoprotein Antibody, clone 8G5F11
      Alternate Names
      • Glycoprotein Vesicular stomatitis Indiana virus
      • Vesicular stomatitis virus glycoprotein
      • VSIV Glycoprotein
Background InformationGlycoprotein from Vesicular stomatitis virus (UniProt: P03522; also known as VSV) is encoded by the G gene (Gene ID: 1489834) in this virus. VSV belongs to the Rhabdoviridae family and two distinct serotypes of this virus have been described the New Jersey and Indiana type. It is an enveloped, negative-sense RNA virus that mainly infects a variety domestic animals and causes vesiculation, ulceration, and erosion of the oral and nasal mucosa and epithelial surface of the tongue. The genomic structure of this virus is a single strand of negative-sense RNA that is composed of five genes known as N, P, M, G, and L that represent the nucleocapsid protein, phosphoprotein, matrix protein, glycoprotein, and the large protein, which is a component of the viral RNA polymerase. The virus glycoprotein is a single pass type 1 membrane protein that attaches virus to host LDL receptors and induces clathrin-dependent endocytosis of the virion. In the endosome, the acidic pH induces conformational changes in the glycoprotein trimer that triggers fusion between virus and endosomal membrane. Because the VSV-Glycoprotein can bind to numerous cell types, it is used for the transduction of genetic material into cells as a vaccine vector and as an antitumor agent. The glycoprotein consists of a virion surface domain (aa 17-467), a helical domain (aa 468-488), and intra-virion domain (aa 489-511). Clone 8G5G11 is shown to cross-react with and cross-neutralize a variety of vesiculovirus members. The DKD residues (aa 257-259) of glycoprotein are shown to dictate binding of this antibody. Clone 8G5F11 is reported to block infection after endocytosis and before genome reverse transcription. (Ref.: Munis, AM., et al. (2018). J. Virol. 92(23); e00900-18).
