      OverviewConcentrate of Dithiothreitol (DTT; Cat. No. 233155) in phosphate buffer, pH 6.5-7.5. Intended for the isolation of epithelial cells, pathogenic or saprophytic bacteria, fungi, and yeasts from sputum. Each vial is sufficient to make 100 ml of working solution.

      DescriptionA concentrate of dithiothreitol (DTT) in phosphate buffer. Useful for the isolation of pathogenic or saprophytic bacteria, fungi, yeast, and epithelial cells from sputum and other mucoid fluids.

      Formulation20 vials each containing 10 ml of concentrated DTT OR 1 vial containing 10 ml of concentrated DTT.
      Recommended reaction conditionsA. To prepare sputum samples: 1. In a screw-cap centrifuge tube, overlay the sputum sample with an equal volume of the diluted SPUTOLYSIN® Reagent. 2. Suspend the sputum by mixing on a vortex mixer for 30 s. 3. Allow the suspended mixture to stand at room temperature for 15 min. 4. Centrifuge the mixture for 5 min at 1500 rpm. Discard the supernatant. B. To prepare cultures for acid-fast bacilli: 1. Suspend the pellet from Step A(4) in 5 to 10 ml of 1% sodium hydroxide by mixing on a vortex mixer for 1 min. 2. Centrifuge the mixture for 15 min at 3000 rpm. Discard the supernatant. 3. Rinse the pellet twice in 10 ml of sterile normal saline to remove the residual sodium hydroxide. 4. Resuspend the pellet in 0.5 ml of the diluted SPUTOLYSIN® Reagent.

      SolubilityDilute the contents of one vial to 100 ml with sterile distilled water. Swirl gently to dissolve any crystals. If the diluted material appears cloudy, the solution may be filtered to remove any insoluble particles. The diluted solution is ~6.5 mM dithiothreitol in 100 mM phosphate buffer, pH 7.0.
