Background InformationTumor necrosis factor ligand superfamily member 15 (UniProt: Q5UBV8; also known as TNF ligand-related molecule 1, TL1A, Vascular endothelial cell growth inhibitor) is encoded by the Tnfsf15 (also known as Tl1, Vegi) gene (Gene ID: 326623) in murine species. TNFSF15 is a single-pass type II membrane protein of the tumor necrosis factor family that is expressed on activated macrophages, dendritic cells, T cells, and endothelial cells and is produced either as a membrane-anchored protein or a soluble cytokine. TNFSF15 has a cytoplasmic domain (aa 1-39), a transmembrane domain (40-60), and an extracellular domain (aa 61-252). Its expression is increased in macrophages and dendritic cells in response to various stimuli. It serves as a ligand for death receptor 3 (DR3) and mediates the activation of NF-kappa-B. It is a key player in the regulation of T-cell-mediated immune responses and promotes T cell immunity to pathogens and directly co-stimulates group 2 and 3 innate lymphoid cells. It is also shown to enhance the proliferation of in vitro generated Th17 cells but does not affect the proliferation of Th1 cells.
