Background InformationThe Rab family of proteins are a member of the Ras superfamily of small G-Proteins. At least 60 isoforms have been identified. These GTPases are primarily involved in regulating distinct steps in vesicle trafficking by regulating the dynamic assembly and disassembly of the multi-protein scaffolds that are involved in vesicle trafficking. In its active, GTP-bound state, the Rab protein recruits effector proteins to the membrane. Rabs lack intrinsic GTPase hydrolysis activity and thus rely on guanine nucleotide exchange factors (GEFs) and GTPase activating proteins (GAPs) to regulate their activity. Through these effectors, they regulate vessicle formation and movement. Rab4 and Rab11 are involved in internalization, sorting and transport to lysosomes or recycling of several membrane proteins, including GPCRs, GLUT-4, Aquaporin-2 or the H+/K+ ATPase.

UniProt SummaryFunction: Probable substrate-recognition component of a SCF-like ECS (Elongin-Cullin-SOCS-box protein) E3 ubiquitin ligase complex which mediates the ubiquitination and subsequent proteasomal degradation of target proteins.
Pathway: Protein modification; protein ubiquitination.
Subunit structure: Component of the probable SCF-like ECS(RAB40C) E3 ubiquitin-protein ligase complex which contains CUL5, RNF7/RBX2, Elongin BC complex and RAB40C. Interacts with CUL5, RNF7, TCEB1 and TCEB2.
Subcellular location: Cell membrane; Lipid-anchor; Cytoplasmic sidePotential.
Domain: The SOCS box domain mediates the interaction with the Elongin BC complex, an adapter module in different E3 ubiquitin ligase complexes By similarity.
Sequence similarities: Belongs to the small GTPase superfamily. Rab family.
Contains 1 SOCS box domain
Molecular Weight31 kDa
